I think I will give three top Varminter cartridges with which I have been so impressed with in the past. Here goes 22-250 Ackley Improved, 6mm BR and the 243 Ackley Improved. Properly chambered (throated) these will perform extremely well and the brass will last a long time. In addition with the components and care you are obviously putting into this rifle the inherent accuracy of these cartridges will make up for any small loss in velocity say in choosing the 243 Ackley as opposed to the 6mm Ackley (of which I own and love one of). I have confidence in my custom 6mm BR and would not hesitate to try a 650 yard shot with it from a good rest. I have a Varminting buddy that built a 243 Ackley improved more than 20 years ago. He bought some RWS brass for it then and did load development and has fired his brass at least once each year since then while Varminting. The brass is still fine! No failures as yet. My 240 Weatherby is a brass eater compared to that and not much (if any) improvement in velocity. So I NOW choose my Varminters with a strong consideration toward quality of available brass (6mm BR by Norma and LaPua). Someone I believe either Norma or LaPua is making 243 Brass. I believe. I know Norma used to anyway. I also have two 22 BR that are both extremely accurate. I think the 22 BR would be my fourth choice for you in case you do not have a 22-250 (though I would move that choice up to second if you do not have a 22-250 Rem.). Good luck with whichever you choose and be sure and get a good set of Redding dies and the BR Seater.
By the way I only have one tight necked Varminter and I am not sure that your accuracy would suffer much at all by using a minimum standard chamber.
